A 2025 structural biology study determined cryo-EM structures of the purified dopamine transporter (DAT) protein in complex with Tesofensine and four other triple reuptake inhibitors. In this in vitro system, Tesofensine stabilized DAT in an outward-facing conformation. The study characterized binding poses and molecular interactions at the transporter [Li et al., 2025].
