VIP-expressing interneurons (VIP-INs) are a focus of active neuroscience research. In a study using pathway-specific electrophysiology with optogenetic and chemogenetic manipulations in mouse hippocampal CA1, Iloun et al. (2026) reported that VIP-INs provide direct inhibitory synaptic input to pyramidal neurons in both superficial and deep sublayers. Chemogenetic silencing of VIP-INs uncovered sublayer- and pathway-specific disinhibitory motifs: during Schaffer collateral activation, VIP-IN silencing preferentially reduced inhibition onto deep pyramidal cells, while during temporoammonic input processing, distinct sublayer-specific effects on excitatory and inhibitory drive were observed. These findings characterize VIP-INs as pathway- and sublayer-specific modulators of excitation-inhibition balance in the mouse hippocampus [1].
